樣品采集和存儲

請注意您感興趣的套件推薦哪些樣品類型。

血清

使用血清分離管，讓樣品在室溫下凝固2小時或在4℃下過夜，然后以約1000×g離心20分鐘。立即測定新鮮制備的血清，或?qū)悠贩盅b在-20℃或-80℃下備用。避免反復(fù)凍/融循環(huán)。

等離子體

使用EDTA或肝素作為抗凝血劑收集血漿。在收集后30分鐘內(nèi)，在2-8℃下以1000×g離心樣品15分鐘。立即去除血漿并進(jìn)行檢測，或?qū)悠繁４嬖?20℃或-80℃的等分試樣中以備后用。避免反復(fù)凍/融循環(huán)。

組織勻漿

組織勻漿的制備將根據(jù)組織類型而變化。通常，組織應(yīng)在冰冷的PBS（0.02mol / L，pH 7.0-7.2）中*沖洗以除去過量的血液并在均質(zhì)化前稱重。將組織切成小塊并在冰上用玻璃勻漿器在5-10mL PBS中均化（Micro Tissue Grinders也起作用）。應(yīng)使用超聲波細(xì)胞破碎器對所得懸浮液進(jìn)行超聲處理，或進(jìn)行兩次凍融循環(huán)以進(jìn)一步破壞細(xì)胞膜。之后，勻漿應(yīng)以5000×g離心5分鐘。立即取出上清液并進(jìn)行分析，或等分試樣并在≤-20℃下儲存。

細(xì)胞裂解物

根據(jù)以下說明，在測定前必須裂解細(xì)胞：

應(yīng)用胰蛋白酶分離貼壁細(xì)胞，然后通過離心收集（懸浮細(xì)胞可通過直接離心收集）。
在冷PBS中洗滌細(xì)胞三次。
在1X PBS中重懸細(xì)胞并進(jìn)行4次超聲波處理（或在≤-20℃下冷凍細(xì)胞。輕輕混合解凍細(xì)胞。重復(fù)凍融循環(huán)3次。）

唾液

尿

注意：

樣品在5天內(nèi)可以被存儲在4使用Ô C，否則樣品必須儲存在-20 Ô C（≤1月）或-80 Ô C（≤2個月），以避免生物活性和/或污染的損失。
樣品溶血會影響結(jié)果; 不會檢測到用作樣本的溶血標(biāo)本。
進(jìn)行測定時，將樣品置于室溫下。

試劑準(zhǔn)備

*使用前將所有試劑盒組分和樣品置于室溫（18-25 o C）。

標(biāo)準(zhǔn)

用標(biāo)準(zhǔn)稀釋劑重新配制標(biāo)準(zhǔn)品，在室溫下保持10分鐘，輕輕搖動（不要起泡）。有關(guān)儲備溶液中標(biāo)準(zhǔn)濃度的信息，請參閱手冊。準(zhǔn)備7管含有0.5mL標(biāo)準(zhǔn)稀釋劑的試管，并使用稀釋的標(biāo)準(zhǔn)品生產(chǎn)雙稀釋系列。在下次轉(zhuǎn)移之前*混合每個管。

檢測試劑A和檢測試劑B.

傳統(tǒng)的ELISA試劑盒：使用前短暫旋轉(zhuǎn)或離心檢測A和檢測B溶液。分別用Assay Diluent A和B稀釋至工作濃度（1：100）。

即用型ELISA試劑盒：檢測溶液A和B的濃度已經(jīng)正確，無需進(jìn)一步稀釋。

洗液

用580mL去離子水或蒸餾水稀釋20mL洗滌液濃縮物（30×），制備600mL洗滌液（1×）。

TMB底物

用滅菌的吸頭吸取所需劑量的溶液。不要將殘留的溶液倒回到小瓶中。

注意：

不要直接在孔中進(jìn)行連續(xù)稀釋。
在進(jìn)行測定后15分鐘內(nèi)制備標(biāo)準(zhǔn)品。不要直接在37 o C 溶解試劑。
檢測試劑A和B是粘性溶液，因此慢慢移液它們以減少體積誤差。
傳統(tǒng)的ELISA試劑盒：根據(jù)說明小心地重新構(gòu)建標(biāo)準(zhǔn)品或使用檢測試劑A和B，避免起泡并輕輕混合直至晶體*溶解。為了大限度地減少移液造成的不性，請使用小體積并確保移液器已校準(zhǔn)。建議一次移液10μL以上以確保準(zhǔn)確性。
即用型ELISA試劑盒：重組標(biāo)準(zhǔn)品只能使用一次。
重構(gòu)的標(biāo)準(zhǔn)品，檢測試劑A和檢測試劑B只能使用一次。
如果在洗滌液濃縮物（30×）中形成晶體，則溫?zé)嶂潦覝夭⑤p輕混合直至晶體*溶解。
試劑制備過程中使用的任何污染水或容器都會影響檢測結(jié)果。

樣品制備

Reddot Biotech僅對試劑盒本身負(fù)責(zé)，而不是對試驗(yàn)過程中消耗的樣品負(fù)責(zé)。實(shí)驗(yàn)者應(yīng)該在開始實(shí)驗(yàn)之前總是計(jì)算所需的樣品總量。
實(shí)驗(yàn)者還應(yīng)該在開始之前預(yù)測大致的濃度。如果預(yù)期值不在標(biāo)準(zhǔn)曲線范圍內(nèi)，建議確定樣品的宜稀釋度。
如果您使用的樣品類型未在手冊中指出，則應(yīng)在開始實(shí)際實(shí)驗(yàn)之前進(jìn)行初步實(shí)驗(yàn)以確保試劑盒的有效性。
建議不要使用化學(xué)裂解緩沖液來制備組織或細(xì)胞提取樣品，因?yàn)樗赡軙捎诹呀饩彌_液的含量而導(dǎo)致意外的反應(yīng)和結(jié)果。
由于其他來源的抗原與我們試劑盒中使用的抗體之間可能存在錯配，因此不建議使用其他制造商生產(chǎn)的天然或重組蛋白，因?yàn)槲覀兊脑噭┖锌赡軣o法識別它們。
我們的試劑盒可能無法檢測到細(xì)胞培養(yǎng)上清液中的樣品，因?yàn)樗鼈兒苋菀资艿郊?xì)胞活力，細(xì)胞數(shù)量和/或取樣時間等因素的影響。
建議將未經(jīng)長期儲存的新鮮樣品與我們的試劑盒一起使用。如果樣品長時間未冷凍保存，可能會發(fā)生蛋白質(zhì)降解和變性，導(dǎo)致結(jié)果相互矛盾或不正確。

分析程序

用于夾心ELISA試劑盒

確定稀釋標(biāo)準(zhǔn)品，空白樣品和樣品的孔。為標(biāo)準(zhǔn)準(zhǔn)備7口井，為空白準(zhǔn)備1口井。每種稀釋標(biāo)準(zhǔn)品（讀取試劑制備），空白和樣品加入100μL到適當(dāng)?shù)目字?。用Plate密封劑蓋住。在37 o C 孵育2小時。

注意：

樣品或試劑添加：請使用新制備的標(biāo)準(zhǔn)品。小心地將樣品加入孔中并輕輕混合以避免起泡。不要觸摸井壁。對于該程序中的每個步驟，將試劑或樣品添加到測定板的總分配時間不應(yīng)超過10分鐘。這將確保每個移液步驟的相等的經(jīng)過時間，而不會中斷。建議復(fù)制所有標(biāo)準(zhǔn)品和標(biāo)本，盡管不是必需的。為避免交叉污染，在添加標(biāo)準(zhǔn)品，樣品和試劑之間更換移液器吸頭。此外，對每種試劑使用分離的儲庫。
孵育：為了確保準(zhǔn)確的結(jié)果，需要在孵育步驟中適當(dāng)粘附板密封劑。在孵化步驟之間不允許孔長時間未被覆蓋。一旦將試劑添加到孔條中，在測定期間不要讓條帶干燥。必須控制孵育時間和溫度。
洗滌：洗滌程序至關(guān)重要。在每個步驟中*去除液體對于試劑盒的良好性能是*的。后一次清洗后，通過抽吸或傾析除去任何殘留的清洗液，并去除板底部的任何水滴或指紋。洗滌不足將導(dǎo)致精度差和吸光度讀數(shù)錯誤升高。
控制反應(yīng)時間：觀察加入TMB底物后的顏色變化（例如每10分鐘觀察一次），如果顏色太深，則提前加入終止溶液以避免過強(qiáng)的反應(yīng)，這將導(dǎo)致吸光度讀數(shù)不準(zhǔn)確。
TMB底物 很容易被污染。保護(hù)它免受光線和物理污染。
環(huán)境濕度可能對從試劑盒獲得的結(jié)果產(chǎn)生影響。如果您工廠的濕度低于60％，建議使用加濕器。

測試原理

用于夾心ELISA試劑盒

靈敏度

確定所有套件精度的一般方法如下所述。可根據(jù)要求提供個人批量套件的規(guī)格。

測定內(nèi)精密度（測定內(nèi)的度）：分別在一個平板上測試3個具有低，中和高水平指數(shù)的樣品20次。
批間精密度（測定之間的度）：在3個不同的平板上測試3個具有低，中和高水平指數(shù)的樣品，每個平板8個重復(fù)。

穩(wěn)定性

當(dāng)次打開板時，孔中可能存在霧狀物質(zhì)。它對終的測定結(jié)果沒有任何影響。在需要之前，不要從儲存袋中取出微量滴定板。
試劑準(zhǔn)備和裝載過程中的程序不正確，以及讀板器的參數(shù)設(shè)置不正確可能導(dǎo)致錯誤的結(jié)果。在450±10nm波長下帶寬為10nm或更小且光密度范圍為0-3OD或更大的微孔板讀數(shù)器可用于吸光度測量。請仔細(xì)閱讀說明書并在實(shí)驗(yàn)前調(diào)整儀器。

該試劑盒中的標(biāo)準(zhǔn)品以及抗體制備中使用的抗原通常是重組蛋白質(zhì)。不同表達(dá)的序列，表達(dá)系統(tǒng)和/或純化方法可用于制備重組蛋白。在我們的試劑盒中，抗體和抗體對的篩選技術(shù)也存在差異。因此，我們無法保證我們的試劑盒能夠檢測其他公司生產(chǎn)的重組蛋白。我們不建議使用Reddot Biotech ELISA試劑盒檢測其他重組蛋白。
有效期：12個月。
說明手冊也適用于48T套件，但48T套件中的所有試劑都減少了一半。
